Response: Hexachlorobenzene

چکیده

There is no PF for TCDD currently available in the Integrated Risk Information Service database (3). However, the PF originally derived from the aforementioned study was 156,000 mg/kg/day and may be modified downward to approximately 100,000 mg/kg/day, based on reinterpretation of histopathologic data. Applying van Birgelen's TEF to these values would suggest a PF for the carcinogenic action of HCB (under the linearized multistage assumptions) to be approximately 10-16 mg/kg/day, or about an order of magnitude more potent than is suggested by the in vivo data. It is possible that the difference lies in pharmacokinetic or pharmacodynamic factors that exist in the whole animal, which cannot be captured in cellular assays. As van Birgelen points out (1), the World Health Organization indicates a preference for long-term in vivo studies over in vitro measures when setting a TEF (5). The HCB example illustrates the extreme caution that should be exercised when applying a TEF based solely on in vitro information, particularly when major public health claims are being made.

Dispersion Coefficients of Supercritical Fluid in Fixed Beds

The axial dispersion coefficient of hexachlorobenzene in supercritical carbon dioxide is investigated in a fixed-bed packed with glass beads. The on-line chromatographic pulse-response experiment is used in order to study the dynamics of a packed column under supercritical conditions. The radial dispersion is assumed negligible because of the packed column geometry. To estimate the axial disper...

Figure 6-1. Frequency of NPL Sites with Hexachlorobenzene Contamination

Hexachlorobenzene has not been manufactured in the United States (as an end-product) since its last registered use as a pesticide was voluntarily canceled in 1984. Small amounts of hexachlorobenzene are still released to the environment by industrial activities as fugitive and stack emissions, or in waste water. It is released as a by-product during the manufacture of several chlorinated solven...

Hexachlorobenzene treatment on hepatic mitochondrial function parameters and intracellular coproporphyrinogen oxidase location.

These studies try to elucidate why isocoproporphyrin appears in hexachlorobenzene-poisoned rats' feces. Chronic exposure of hexachlorobenzene to rats produces an experimental model for human porphyria cutanea tarda. After 8 weeks of treatment, rats showed high porphyrin excreta and 50% inhibition of liver uroporphyrinogen decarboxylase activity. Uroporphyrin plus heptacarboxylic porphyrin excee...

Reductive dechlorination of a polychlorinated biphenyl congener and hexachlorobenzene by vitamin B12.

The polychlorinated biphenyl congener 2,3,4,5,6-pentachlorobiphenyl and hexachlorobenzene were reductively dechlorinated in an aqueous biomimetic model system containing vitamin B12. The products of 2,3,4,5,6-pentachlorobiphenyl dechlorination were 2,3,5,6- and 2,3,4,6-tetrachlorobiphenyl. Hexachlorobenzene dechlorinated to pentachlorobenzene and a mixture of 1,2,4,5- and 1,2,3,5-tetrachloroben...

The effect of the porphyrogenic compound, hexachlorobenzene, on the activity of hepatic uroporphyrinogen decarboxylase in the rat.

1. A new method for the measurement of uroporphyrinogen decarboxylase (EC 4. 1.1.37) in rat liver homogenates, with 5- carboxyl porphyrinogen as substrate, is described. 2. The administration of a diet containing 0-3% (w/w) hexachlorobenzene produces porphyria in female Wistar rats after a delay of at least 4 weeks. The development of porphyria is accompanied by a progressive fall in hepatic ur...

Testing of p-dichlorobenzene and hexachlorobenzene for their ability to induce DNA damage and micronucleus formation in primary cultures of rat and human hepatocytes.

The genotoxicity of p-dichlorobenzene (DCB) and hexachlorobenzene (HCB) was evaluated in primary cultures of rat and human hepatocytes. DNA fragmentation was measured by the alkaline elution technique and clastogenic activity by the increase in micronucleus formation. In rat hepatocytes, exposure to concentrations of DCB ranging from 0.56 to 3.2 mM and of HCB from 0.1 to 0.56 mM did not induce ...
